With more than 4500 km, it starts in the Himalayas and crosses the entire region until it reaches Vietnam in the famous Mekong Delta.
You can go by your own but for this type of adventure, I recommend to take a tour. There are tours for spending 1 or 2 nights close to the Mekong River.
The ideal experience would be to visit some of the towns in the area, floating markets, and rice fields.

The Mekong river is one of the largest most well known rivers in south east Asia, stretching from Tibet and ending at the Delta in Vietnam. Come with a coconut in hand and experience the traditional ways of the local Vietnamese that live by the river to see their traditional way of life.

For a more rural experience, go to the Mekong Delta and take a boat ride to see how Vietnamese have lived for years. They live off of the river and farm nearby. Rice is the largest crop in the region and they use every bit of it from making rice paper, rice candies, rice wine, and of course rice the grain. The locals are friendly and hospitable. There are boats you can buy fresh fruit and goods from in the floating market. Great experience to see how other cultures operate.

My travel style is more of like a DIY-style, so when I took a day tour here, it was ok but it didn't make an impact to me at all. I was looking forward to the mangroves but we ride the boat for only 10 minutes.

Quite a journey to get there. If possible try to arrange a small group trip and not mass visit.

The Mekong Delta is a beautiful tourist attraction, as we got to travel by the seas and visit Unicorn and Dragon Island. See where the coconuts are harvested and made into candy! In small boats, we sailed down the Mekong River in pouring rain, giving new meaning to adventures in the high seas!

the tour was ok but nothing memorable. unfortunatelly did not see much of the floating market, but the village production on several rice products ( wine etc) was interesting.

The quiet sounds of river streams and women in cone hats rowing long boats with painted eyes (used for good luck and to ward off bad spirits) along the river makes the Mekong Delta an enticing trip. If you want to get a more localized taste of Vietnam, then a trip to the Delta is highly advised.

I did a tour with buffalo tours and was the only person. The Mekong delta is great for pictures. It's really active and different from the floating markets you nights see in Thailand. This feels more like the pulse and lifeline and your pics will reflect that.

If I can only recommend one tour to do while in Ho Chi Minh, it would be the Mekong Delta Discovery Trip. This can be arranged in any of the tour agencies in the city and your best bet would be from any of the agencies along Pham Ngu Lau, Saigon’s backpacker district. This tour is a great introduction to the mighty Mekong River which ripples past rice fields, blossoming fruit orchards and towering sugarcane fields before tumbling into the South China Sea. The air-conditioned bus will take you to My Tho City and you will visit two of the four islands in the area. You will wind your way along the beautiful little canals under a canopy of coconut trees.
The tour also includes a stop at a small village to visit a bee farm where you get to sample a concoction of honey and tea, enjoy a plate of tropical fruits, learn about the way of life of the islanders, see them working at various forms of agriculture, and be entertained by their traditional music. The best part, for me, is that we were able to paddle a row boat while navigating through a mangrove forest area after two ferry crossings, which is what this little adventure was about. It would be a wise idea to bring a wide-brimmed hat-- essential protection from the sun, and even carry an umbrella to be used either as a parasol or as cover from sporadic rains. Bring a pair of good sandals as well!
